Effects Of Crotalus durissus collilineatus Venom In The Isolated Rat Kidney.

Crotalusdurissus collilineatus (Cdco) is a snake usually found in semideciduous forest, the Cerrado region and its bites constitutes an important public health problem.The most serious systemic change and primary cause of death is acute renal failure although the mecanisms of the damaging effects are not totally understood. In this work, we have examined the renal effects caused by Cdco venom in the isolated rat kidney perfused with Krebs-Henseleit solution containing 6% of bovine serum albumin.The parameters studied included perfusion pressure (PP), glomerular filtration rate (GFR), urinary flow (UF),renal vascular resistance (RVR), percent of sodium tubular transport (%TNa+), percent of potassium tubular transport (%TK+), and percent of chloride tubular transport (%TCl-). After an equilibration period of 20min, the experiments were carried out for 120min. Samples of perfusate were collected every 10min, for determination of sodium, potassium,and chloride.The UF rate was also measured at 10min intervals. The data were analyzed by Student's t-test with the level of significance of p<0,05.The infusion of Cdco (10mg/mL) after 30min of an internal control caused a significantly increase in PP(mmHg), GFR(mL/g/min) and RVR(mmHg/mL/g/min) with maximal effect at 120min of perfusion: PP (control120

110,3 ± 3,7; venom120 126,8±10,2), GFR (control120 0,69±0,08; venom120 1,17±0,39) and RVR (control120 5,20±0,5; venom120 6,67±0,94). The UF(mL/g/min) also significantly increased at 120min: control120 0,16 ± 0,02; venom120 0,23 ± 0,06.The %TCl- remained stable during the 120min of perfusion.These results indicate that Crotalusdurissus collilineatus venom caused significative alterations in the renal parameters.
